What is quercetin?
A primary limitation in the formulation development and therapeutic application of quercetin is its poor systemic bioavailability. Due to its highly lipophilic nature, quercetin exhibits negligible aqueous solubility, resulting in poor dissolution within gastrointestinal fluids and limited intestinal absorption following oral administration.
The compound also undergoes rapid chemical degradation under physiological conditions, alongside extensive first-pass hepatic metabolism and intestinal glucuronidation. Coupled with low membrane permeability and a rapid systemic clearance rate, these pharmacokinetic challenges culminate in low, highly variable circulating plasma concentrations of the active compound.
